How Pregnancy Tests Work: The 3-Minute Chemistry Inside the Strip
Sandwich antibodies, dye nanoparticles and capillary action — the elegant lateral-flow machine inside every test, explained so every confusing result finally makes sense.

Every confusing result — faints, evaps, hooks, dye stealers — becomes obvious once you know what's physically happening on the strip. Here's the machine, in the order events unfold.
The cast
A test strip is four zones on a nitrocellulose membrane: a sample pad where urine lands; a conjugate pad holding dried antibodies tagged with colored nanoparticles (classically gold — pink!); a test stripe of fixed anti-hCG antibodies; and a control stripe that catches tagged antibodies regardless of hCG. Capillary action is the only motor.
The four acts
- Wetting. Urine soaks the sample pad and dissolves the tagged antibodies; the pink wave begins moving — the flow you can watch in the window.
- Binding. If hCG is present, tagged antibodies grab it mid-flow — each hormone molecule now wears a colored tag on one end.
- The sandwich. At the test stripe, fixed antibodies grab the other end of any passing hCG. Hormone gets pinned with its tag — pink accumulates — a line forms. No hCG, nothing pins, no line. Line intensity tracks how much hormone flowed past, which is all of progression in one sentence.
- The control. Leftover tagged antibodies flow on and are captured at the control stripe by species-specific antibodies — proving flow and reagents worked. No control, no verdict.
Every mystery, explained by the machine
- Faint lines = little hormone, few sandwiches — early days or dilute urine.
- Sensitivity = how little hCG still builds a visible pink stripe.
- Evaps = drying residue on the stripe's topography — chemistry finished, physics doodling.
- Indents = the printed stripe itself, visible without any dye.
- Dye stealers = so much hCG the test stripe exhausts tags before the control gets any.
- The hook = extreme hCG saturating both antibodies separately, so no sandwich completes.
- One-time use = the conjugate pad's tags all left in the first flow.

Why it's a great design
